Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
The peer-reviewed record is dominated by small early-phase studies, case reports and pharmacovigilance summaries rather than large randomised trials. Papers typically examine tanning response, receptor selectivity or patterns of reported adverse events. Many note that participants obtained the peptide outside a clinical setting, which limits verification of composition and administered amount. Reported events vary widely, and causality is frequently unclear because the identity and purity of self-sourced material are unknown. Open questions include whether repeated melanocortin receptor stimulation produces cumulative effects, and how often label claims match actual content.
Identification in laboratories relies on reversed-phase liquid chromatography coupled with tandem mass spectrometry, with product-ion spectra compared against a certified reference standard. High-resolution mass spectrometry supplies accurate mass confirmation, and peptide mapping after enzymatic digestion separates melanotan II from closely related analogues. Quantitation of seized material is complicated by unknown counter-ions and residual trifluoroacetate left from purification. Immunoassays raised against alpha-melanocyte-stimulating hormone can cross-react, so chromatographic confirmation is normally required. Urinary detection windows are short, and reported limits of detection differ substantially between laboratories.

Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.

Root: It is the attached part, consisting of the bulb in the middle and the crura, one crus on either side of the bulb. It lies within the superficial perineal pouch. The crus is attached to the pubic arch. Shaft: The pendulous part of the penis. It has two surfaces: dorsal (posterosuperior in the erect penis) and ventral or urethral (facing downwards and backwards on the flaccid penis). The ventral surface is marked by the penile raphe. The base of the shaft is supported by the suspensory ligament, which is attached to the pubic symphysis. Epithelium of the penis consists of the shaft skin, the foreskin (prepuce), and the preputial mucosa on the inside of it. The foreskin covers and protects the glans and shaft. The epithelium is not attached to the underlying shaft, so it is free to glide to and fro. The human penis is made up of three columns of erectile tissue: two corpora cavernosa lie next to each other (separated by a fibrous septum) on the dorsal side and one corpus spongiosum lies between them on the ventral side. These columns are surrounded by a fibrous layer of connective tissue called the tunica albuginea. The corpora cavernosa are innervated by lesser and greater cavernous nerves and form most of the penis containing blood vessels that fill with blood to help make an erection. The crura are the proximal parts of the corpora cavernosa. The corpus spongiosum is an erectile tissue surrounding the urethra. The proximal parts of the corpus spongiosum form the bulb and the distal ends form the glans penis.
After details of past social media posts made by two Reform UK candidates, Lee Bunker and Angela Begbie-Carter, are reported in the media, the party releases a statement in which it says candidates are free to express views that "are not shared by all their party colleagues" as they are not "political zombies". Secret government files seen by BBC News show work has been underway with consultants since January to limit "severe implications" to essential IT services by lining up alternative providers after Atos, the UK subsidiary of which has contracts for NHS records and disability benefit claims, is more than £3bn in debt and undergoing restructuring. Sinn Féin launches its 2024 election manifesto, which includes plans for the transfer of fiscal powers from Westminster to Stormont and the creation of an all-Ireland national health service. Businessman Zia Yusuf donates an undisclosed amount of money, but reported to be several thousand pounds, to Reform UK, saying the UK has "lost control of our borders". The Workers Party of Britain launches its election manifesto, with promises to improve "poverty pay" and provide more social housing. BBC News reports that Laura Saunders, the Conservative candidate for Bristol North West, has become the second Conservative candidate to face an investigation by the Gambling Commission over betting on the date of the general election. It is subsequently reported that her husband, Tony Lee, the Conservative Party's campaigns director, is also being investigated by the Commission.
2-Aminoisobutyric acid (Aib) is the non-proteinogenic amino acid with the structural formula H2N-C(CH3)2-COOH. The Aib residue is a component of tirzepatide, a commonly prescribed antidiabetic medication for treatment of type 2 diabetes. Although uncommon, it is also found in some natural products 2-Aminoisobutyric acid can be prepared from acetone cyanohydrin, by reaction with ammonia followed by hydrolysis. The compound is not chiral, unlike most amino acids. It is a strong helix inducer in peptides due to Thorpe–Ingold effect of its gem-dimethyl group. Oligomers of Aib form 310 helices.
The first National Air Pollution Symposium in the United States was held in 1949 and hosted by Stanford Research Institute (now SRI International). At first, smaller governments were responsible for the passage and enforcement of such legislation. The main purpose of the Air Pollution Control Act of 1955 was to provide research assistance to find a way to control air pollution from its source. A total of $5 million was granted to the public health service for a five-year period to conduct this research. According to a private website, the amount was $3 million allotted per year for the five-year period of research.
== Hazards == Americium-241 has the same general hazards as other americium isotopes: it is both extremely toxic and radioactive. Though α-particles can be stopped by a sheet of paper, there are serious health concerns for ingestion of α-emitters. Americium and its isotopes are also very chemically toxic as well, in the form of heavy-metal toxicity. As little as 0.03 microcuries (1.1 kBq) is the maximum permissible body burden for 241Am. Americium-241 is an α-emitter with a weak γ-ray byproduct. Safely handling americium-241 requires knowing and following proper safety precautions, as without them it would be extremely dangerous. Its specific gamma dose constant is 3.14×10−1 mR/hr/mCi or 8.48×10−5 mSv/hr/MBq at 1 metre (3 ft 3 in). If consumed, americium-241 is excreted within a few days and only 0.05% is absorbed in the blood. From there, roughly 45% of it goes to the liver and 45% to the bones, and the remaining 10% is excreted. The uptake to the liver depends on the individual and increases with age. In the bones, americium is first deposited over cortical and trabecular surfaces and slowly redistributes over the bone with time. The biological half-life of 241Am is 50 years in the bones and 20 years in the liver, whereas in the gonads (testicles and ovaries) it remains permanently; in all these organs, americium promotes formation of cancer cells as a result of its radioactivity.

4 On 21 December 1995, a major reshuffle took place amongst existing Ministers, mainly to discharge particular Ministers of portfolios which had proven troublesome for the government. 5 On 26 April 1996, George Cash resigned from the ministry due to ill health. Graham Kierath and Kevin Minson adopted his portfolios, whilst Norman Moore was elected leader of the Government in the Legislative Council four days later.
is large, which results in a larger plastic radius. This implies that the material can plastically deform, and, therefore, is tough. This estimate of the size of the plastic zone beyond the crack tip can then be used to more accurately analyze how a material will behave in the presence of a crack. The same process as described above for a single event loading also applies and to cyclic loading. If a crack is present in a specimen that undergoes cyclic loading, the specimen will plastically deform at the crack tip and delay the crack growth. In the event of an overload or excursion, this model changes slightly to accommodate the sudden increase in stress from that which the material previously experienced. At a sufficiently high load (overload), the crack grows out of the plastic zone that contained it and leaves behind the pocket of the original plastic deformation. Now, assuming that the overload stress is not sufficiently high as to completely fracture the specimen, the crack will undergo further plastic deformation around the new crack tip, enlarging the zone of residual plastic stresses. This process further toughens and prolongs the life of the material because the new plastic zone is larger than what it would be under the usual stress conditions. This allows the material to undergo more cycles of loading. This idea can be illustrated further by the graph of Aluminum with a center crack undergoing overloading events.
In anatomy, a fistula (pl.: fistulas or fistulae ; from Latin fistula 'tube, pipe') is an abnormal connection (i.e. tube) joining two hollow spaces (technically, two epithelialized surfaces), such as blood vessels, intestines, or other hollow organs to each other, often resulting in an abnormal flow of fluid from one space to the other. An anal fistula connects the anal canal to the perianal skin. An anovaginal or rectovaginal fistula is a hole joining the anus or rectum to the vagina. A colovaginal fistula joins the space in the colon to that in the vagina. A urinary tract fistula is an abnormal opening in the urinary tract or an abnormal connection between the urinary tract and another organ. An abnormal communication (i.e. hole or tube) between the bladder and the uterus is called a vesicouterine fistula, while if it is between the bladder and the vagina it is known as a vesicovaginal fistula, and if between the urethra and the vagina: a urethrovaginal fistula. When occurring between two parts of the intestine, it is known as an enteroenteral fistula, between the small intestine and the skin it is known as an enterocutaneous fistula, and between the colon and the skin as a colocutaneous fistula. A fistula can result from an infection, inflammation, injury or surgery. Many result from complications during childbirth. Sometimes a fistula is deliberately surgically created as part of a treatment, for example in the case of an arteriovenous fistula for hemodialysis.
Muscular dystrophy (MD) is a genetically and clinically heterogeneous group of rare neuromuscular diseases that cause progressive weakness and breakdown of skeletal muscles over time. The disorders differ as to which muscles are primarily affected, the degree of weakness, how fast they worsen, and when symptoms begin. Some types are also associated with problems in other organs. Over 30 different disorders are classified as muscular dystrophies. Of those, Duchenne muscular dystrophy (DMD) accounts for approximately 50% of cases and affects males beginning around the age of four. Other relatively common muscular dystrophies include Becker muscular dystrophy, facioscapulohumeral muscular dystrophy, and myotonic dystrophy, whereas limb–girdle muscular dystrophy and congenital muscular dystrophy are themselves groups of several – usually extremely rare – genetic disorders. Muscular dystrophies are caused by mutations in genes, usually those involved in making muscle proteins. The muscle protein dystrophin is in most muscle cells and works to strengthen the muscle fibers and protect them from injury as muscles contract and relax. It links the muscle membrane to the thin muscular filaments within the cell. Dystrophin is an integral part of the muscular structure. An absence of dystrophin can cause impairments: healthy muscle tissue can be replaced by fibrous tissue and fat, causing an inability to generate force. Respiratory and cardiac complications can occur as well. These mutations are either inherited from parents or may occur spontaneously during early development.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
Regulatory treatment varies by country. In the United States, the European Union and Australia it is an unapproved drug and its sale is restricted, while some other jurisdictions list it as prescription-only or controlled. The applicable rules depend on the country of import.
