melanogenesis raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2025-12-16. Anything still debated is marked as such rather than presented as settled.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Scientific discussion of Melanotan-2 spans pharmacology, dermatology, and public-health literature. Laboratory studies examine its receptor binding and cellular effects, while clinical reports describe outcomes observed after unregulated use. These two bodies of work differ in rigour and intent. Peer-reviewed trials of the compound as a medicine are limited, so much of the available information comes from case reports and surveillance data. Authors frequently note the gap between experimental findings and real-world use.
Reported observations after unregulated use include shifts in skin pigmentation and, in some accounts, unintended changes to moles and other lesions. Whether these outcomes are causally linked to the compound, and how often they occur, remain open questions because controlled data are scarce. The absence of standardised dosing and verified product purity complicates interpretation. Researchers have called for better surveillance and analytical characterisation of samples obtained outside regulated channels. Conclusions drawn from anecdotal evidence should be treated as provisional.
Melanotan-2 has not received marketing authorisation from major regulatory agencies for any therapeutic indication. Several jurisdictions classify it as a prescription-only medicine or a controlled substance when supplied for human use. Because approved products do not exist, material sold online usually sits outside pharmaceutical supply chains and formal quality oversight. Regulators have issued public notices describing the compound as unapproved. Enforcement varies, and the legal position differs between countries, which complicates any single general statement about its status.

Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.
Melanotan II is a synthetic cyclic heptapeptide that acts as an agonist at melanocortin receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide involved in pigment production. The analogue carries a lactam bridge that constrains the ring and slows enzymatic breakdown relative to the native hormone. In research literature it appears under several abbreviations, and naming conventions are not fully standardized. Published descriptions usually place it within the broader melanocortin agonist family.
Melanotan-2 appears on the World Anti-Doping Agency prohibited list within the peptide hormone class, and several national regulators treat it as an unapproved prescription substance. Some countries restrict importation or sale for personal use. Because the compound is widely traded as a research chemical, the practical legal picture differs between jurisdictions and shifts over time. Human safety data covering long periods are limited, and whether repeated pigmentation changes carry any lasting risk to melanocytes remains an open question.
Freeze-dried melanotan-2 is normally kept as a desiccated powder at minus twenty degrees Celsius or lower, shielded from light and moisture. Peptides of this size degrade through hydrolysis, oxidation and deamidation, and each pathway accelerates as temperature and water activity rise. Repeated freeze-thaw cycles promote aggregation and loss of material, so aliquoting a stock solution before freezing is standard laboratory practice. Once dissolved, the solution is markedly less stable than the powder. In laboratory work, solutions are generally refrigerated and used within days rather than kept for months.
==== Gene Microarrays ==== Traditionally DNA microarrays use complementary DNA or oligonucleotide probes to analyze messenger RNA (mRNA) from genes of interest. Extracted total RNA serves as a template for complementary DNA (cDNA) that is tagged with fluorescent probes before being allowed to hybridize to the microarray for visualization. For proteases, specific probes for protease genes and their inhibitors have been developed to view expression patterns on the mRNA transcript level. The two platforms currently available for this purpose come from corporate and academic sources. Affymetrix's Hu/Mu ProtIn Microarray uses 516 and 456 probe sets to evaluate human and murine proteases, inhibitors, and interactors respectively. CLIP-CHIP™, developed by the Overall Lab, is a complete protease and inhibitor DNA microarray for all 1561 human and murine proteases, non-proteolytic homologues, and their inhibitors. Both of these tools allow comparison of expression patterns between normal and diseased samples and tissues. Unfortunately, as transcript levels often fail to reflect protein expression levels, gene microarrays are limited in representing protein in samples. In addition, proteases recruited from remote sources like nearby tissues are ignored by these DNA based arrays, reiterating the need for protein based methods to confirm the presence and activity of functional enzymes when transcriptome analysis is performed.

In autoimmune hemolytic anemia, concentrations of HbA1 are undetectable. Administration of prednisolone will allow the HbA1 to be detected. The alternative fructosamine test may be used in these circumstances and it also reflects an average of blood glucose levels over the preceding 2 to 3 weeks. All the major institutions such as the International Expert Committee Report, drawn from the International Diabetes Federation, the European Association for the Study of Diabetes, and the American Diabetes Association, suggest the HbA1c level of 48 mmol/mol (6.5 DCCT %) as a diagnostic level. The Committee Report further states that, when HbA1c testing cannot be done, the fasting and glucose-tolerance tests should be done. Screening for diabetes during pregnancy continues to require fasting and glucose-tolerance measurements for gestational diabetes at 24 to 28 weeks gestation, although glycated hemoglobin may be used for screening at the first prenatal visit.

=== Meat packing and trade === Prior to 1882, the South Island of New Zealand had been experimenting with sowing grass and crossbreeding sheep, which immediately gave their farmers economic potential in the exportation of meat. In 1882, the first successful shipment of sheep carcasses was sent from Port Chalmers in Dunedin, New Zealand, to London. By the 1890s, the frozen meat trade became increasingly more profitable in New Zealand, especially in Canterbury, where 50% of exported sheep carcasses came from in 1900. It was not long before Canterbury meat was known for the highest quality, creating a demand for New Zealand meat around the world. In order to meet this new demand, the farmers improved their feed so sheep could be ready for the slaughter in only seven months. This new method of shipping led to an economic boom in New Zealand by the mid 1890s. In the United States, the Meat Inspection Act of 1891 was put in place in the United States because local butchers felt the refrigerated railcar system was unwholesome. When meat packing began to take off, consumers became nervous about the quality of the meat for consumption. Upton Sinclair's 1906 novel The Jungle brought negative attention to the meat packing industry, by drawing to light unsanitary working conditions and processing of diseased animals. The book caught the attention of President Theodore Roosevelt, and the 1906 Meat Inspection Act was put into place as an amendment to the Meat Inspection Act of 1891. This new act focused on the quality of the meat and environment it is processed in.

Mishima Shokuhin's furikake products include "Yukari" and "Umeko," and four of the aforementioned top 20 products in large bags are "Yukari" and one is "Umeko.
In humans, excessive exposure to UV radiation can result in acute and chronic harmful effects on the eye's dioptric system and retina. The risk is elevated at high altitudes and people living in high latitude areas where snow covers the ground right into early summer and sun positions even at zenith are low, are particularly at risk. Skin, the circadian system, and the immune system can also be affected. The differential effects of various wavelengths of light on the human cornea and skin are sometimes called the "erythemal action spectrum". The action spectrum shows that UVA does not cause immediate reaction, but rather UV begins to cause photokeratitis and skin redness (with lighter skinned individuals being more sensitive) at wavelengths starting near the beginning of the UVB band at 315 nm, and rapidly increasing to 300 nm. The skin and eyes are most sensitive to damage by UV at 265–275 nm, which is in the lower UVC band. At still shorter wavelengths of UV, damage continues to happen, but the overt effects are not as great with so little penetrating the atmosphere. The WHO-standard ultraviolet index is a widely publicized measurement of total strength of UV wavelengths that cause sunburn on human skin, by weighting UV exposure for action spectrum effects at a given time and location. This standard shows that most sunburn happens due to UV at wavelengths near the boundary of the UVA and UVB bands.
== Etymology == The term "oxytocin" derives from the Greek ὀξυτόκιον (oxytokion), based on ὀξύς (oxús), meaning "sharp" or "swift", and τόκος (tókos), meaning "childbirth". The adjective form is "oxytocic", which refers to medicines that stimulate uterine contractions, to speed up the process of childbirth. Colloquially, it has been referred to as the "cuddle hormone," "hug hormone," or "love hormone" because of extensive evidence of its involvement in mating and social behavior. It is involved in mating behavior in animals as low as C. elegans.
Overall, these iron oxides display good magnetic properties, lower toxicity, and high stability against degradation. For example, a Fe3-δO4 core-shell is used as a carrier for drug delivery. The designed magnetic nanoparticle-based structure displayed biocompatibility, the formation of a covalent bond between the carrier and drug, and glutathione-responsive drug release which prevents early drug release and increases bioavailability. Furthermore, the presence of magnetic nanoparticles in this drug delivery method allows for its response to external magnetic fields for functionalization. The combination of superparamagnetic iron oxide (SPIO) and polyethylene glycol (PEG) used as drug carriers for doxorubicin are influenced by external magnetism. In vivo SPIO-PEG-D under a magnetic field leads to greater tumor accumulation of therapeutic elements, shows lower tumor size, and reduces cardiotoxicity and hepatotoxicity in the magnetic field. Doxorubicin is known for being extremely toxic, and SPIO-PEG shows potential for use as a nanoparticle carrier for reduced toxicity in the periphery.

Dinosaur eggs are the organic vessels in which a dinosaur embryo develops. When the first scientifically documented remains of non-avian dinosaurs were being described in England during the 1820s, it was presumed that dinosaurs had laid eggs because they were reptiles. In 1859, the first scientifically documented dinosaur egg fossils were discovered in France by Jean-Jacques Pouech, although they were mistaken for giant bird eggs (birds were not yet recognized as dinosaurs at the time). The first scientifically recognized non-avian dinosaur egg fossils were discovered in 1923 by an American Museum of Natural History crew in Mongolia. Dinosaur eggshell can be studied in thin section and viewed under a microscope. The interior of a dinosaur egg can be studied using CAT scans or by gradually dissolving away the shell with acid. Sometimes the egg preserves the remains of the developing embryo inside. The oldest known dinosaur eggs and embryos are from Massospondylus, which lived during the Early Jurassic, about 190 million years ago.
Meanwhile, Khrushchev broadened Moscow's policy to establish ties with India and other key neutral states. Independence movements in the Third World transformed the post-war order into a more pluralistic world of decolonized African and Middle Eastern nations and of rising nationalism in Asia and Latin America.
=== January === January 13, 2009: South Korea, Taiwan, Japan, United States Global shipping experiences a drop in trade, as exports from South Korea dropping an annualised 30%, with Taiwan and Japan experiencing a 42% and 27% drop respectively. === Fat === In 1949, purified insulin was shown to regenerate fat in diabetics with lipoatrophy. In 1976, after 82 days of consecutive injections into a scar, purified insulin was shown to safely regenerate fat and completely regenerate skin in a non-diabetic. During a high-fat diet, and during hair follicle growth, mature adipocytes (fats) are naturally formed in multiple tissues. Fat tissue has been implicated in the inducement of tissue regeneration. Myofibroblasts are the fibroblast responsible for scar and in 2017 it was found that the regeneration of fat transformed myofibroblasts into adipocytes instead of scar tissue. Scientists also identified bone morphogenetic protein (BMP) signalling as important for myofibroblasts transforming into adipocytes for the purpose of skin and fat regeneration.
== As a drug target == NAMPT has increasingly been researched as a potential drug target, with activators of NAMPT having potential applications as anti-aging drugs, while inhibitors of NAMPT may be useful for the treatment of certain forms of cancer. Because cancer cells utilize increased glycolysis, and because NAD enhances glycolysis, iNAMPT is often amplified in cancer cells. APO866 (FK866) is an experimental drug that inhibits this enzyme. It was tested for treatment of advanced melanoma, cutaneous T-cell lymphoma (CTL), and refractory or relapsed B-chronic lymphocytic leukemia, but was dropped from development due to disappointing efficacy results in trials. However, it has been shown to inhibit epithelial–mesenchymal transition (EMT) and inhibit tumor-associated angiogenesis, and may be useful for other medical indications. Anti-aging biomedical company Calico has licensed the experimental P7C3 analogs involved in enhancing iNAMPT activity. P7C3 compounds have been shown in a number of publications to be beneficial in animal models for age-related neurodegeneration.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
Major regulatory agencies have not approved it for any indication. Some countries permit it only under prescription frameworks, while others classify it as a controlled substance.
