Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
The compound was developed in the late 1980s and 1990s by academic researchers investigating photoprotection. The rationale held that stimulating melanin production might reduce ultraviolet damage to skin and lower skin cancer risk. Early work examined receptor binding, pigment response, and short-term tolerability in small studies. That program did not produce an approved drug, and formal development stalled after early-phase trials. Whether induced pigmentation confers meaningful photoprotection remains an open question.
Outside regulated medicine, melanotan II circulates through online vendors as a research chemical, often marketed for tanning. Products sold this way vary widely in purity, concentration, and labeling accuracy, and independent testing has documented discrepancies. Published reports describe both pigment effects and adverse reactions, including nausea, flushing, and darkening of existing moles. Long-term safety data are sparse, and no large controlled trial has established a risk profile. Questions about cumulative effects on melanocytes remain unresolved in the literature.
Melanotan II is a synthetic peptide analog modeled on alpha-melanocyte-stimulating hormone, a naturally occurring signaling peptide involved in pigmentation. Its structure is a cyclic heptapeptide containing two non-natural substitutions, norleucine at position four and D-phenylalanine at position seven. These modifications resist enzymatic breakdown and extend the molecule's activity relative to the native hormone. The compound binds melanocortin receptors and is studied mainly as a pharmacological tool rather than a therapeutic product. It has never received approval as a medicine in any major jurisdiction.

Regulatory treatment of this peptide varies by country. It holds no marketing authorization as a medicine in the United States, the European Union, or most other jurisdictions. Some countries classify products containing it as prescription-only or unlicensed medicines, which restricts lawful supply. Authorities have issued public notices warning that unregulated products may contain undeclared or incorrect ingredients. The molecule also appears on prohibited lists for competitive sport. These measures address supply oversight rather than any approved therapeutic role.
Published research on the compound remains limited. Much of the human data comes from small, early-stage studies rather than large controlled trials, and several questions about effects and variability between individuals remain open. Investigators have examined receptor activity, pigment pathways, and related physiological responses in laboratory and animal models. Findings from those models do not automatically translate to human outcomes. Reviews frequently note the scarcity of rigorous clinical evidence and call for better-characterized study material.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan II holds no marketing authorisation from the Food and Drug Administration, the European Medicines Agency, the UK Medicines and Healthcare products Regulatory Agency or Australia's Therapeutic Goods Administration. Products sold under that name are treated as unapproved new drugs, and their sale or import is prohibited in several jurisdictions. Other countries classify the peptide as a prescription-only medicine or place it among controlled substances, so the legal position changes with the destination market. No pharmacopoeial monograph supplies an official specification, because the material is not a licensed pharmaceutical. Consequently, products offered online are not manufactured to a shared public standard.
The peer-reviewed record is dominated by small early-phase studies, case reports and pharmacovigilance summaries rather than large randomised trials. Papers typically examine tanning response, receptor selectivity or patterns of reported adverse events. Many note that participants obtained the peptide outside a clinical setting, which limits verification of composition and administered amount. Reported events vary widely, and causality is frequently unclear because the identity and purity of self-sourced material are unknown. Open questions include whether repeated melanocortin receptor stimulation produces cumulative effects, and how often label claims match actual content.
Eduard Buchner submitted his first paper on the study of yeast extracts in 1897. In a series of experiments at the University of Berlin, he found that sugar was fermented by yeast extracts even when there were no living yeast cells in the mixture. He named the enzyme that brought about the fermentation of sucrose "zymase". In 1907, he received the Nobel Prize in Chemistry for "his discovery of cell-free fermentation". Following Buchner's example, enzymes are usually named according to the reaction they carry out: the suffix -ase is combined with the name of the substrate (e.g., lactase is the enzyme that cleaves lactose) or to the type of reaction (e.g., DNA polymerase forms DNA polymers). The biochemical identity of enzymes was still unknown in the early 1900s. Many scientists observed that enzymatic activity was associated with proteins, but others (such as Nobel laureate Richard Willstätter) argued that proteins were merely carriers for the true enzymes and that proteins per se were incapable of catalysis. In 1926, James B. Sumner showed that the enzyme urease was a pure protein and crystallized it; he did likewise for the enzyme catalase in 1937. The conclusion that pure proteins can be enzymes was definitively demonstrated by John Howard Northrop and Wendell Meredith Stanley, who worked on the digestive enzymes pepsin (1930), trypsin and chymotrypsin. These three scientists were awarded the 1946 Nobel Prize in Chemistry. The discovery that enzymes could be crystallized eventually allowed their structures to be solved by x-ray crystallography.
However, despite the greater in vitro activity of some of its metabolites, it has been determined that oxycodone itself is responsible for 83.0% and 94.8% of its analgesic effect following oral and intravenous administration, respectively. Oxymorphone plays only a minor role, being responsible for 15.8% and 4.5% of the analgesic effect of oxycodone after oral and intravenous administration, respectively. Although the CYP2D6 genotype and the route of administration result in differential rates of oxymorphone formation, the unchanged parent compound remains the major contributor to the overall analgesic effect of oxycodone. In contrast to oxycodone and oxymorphone, noroxycodone and noroxymorphone, while also potent MOR agonists, poorly cross the blood–brain barrier into the central nervous system, and for this reason are only minimally analgesic in comparison.
Efficacy was based on DESTINY-Breast03 (NCT03529110), a multicenter, open-label, randomized trial that enrolled 524 participants with HER2-positive, unresectable, and/or metastatic breast cancer who received prior trastuzumab and taxane therapy for metastatic disease or developed disease recurrence during or within six months of completing neoadjuvant or adjuvant therapy. Participants were randomized 1:1 to receive either trastuzumab deruxtecan or trastuzumab emtansine by intravenous infusion every three weeks until unacceptable toxicity or disease progression. Randomization was stratified by hormone receptor status, prior treatment with pertuzumab, and history of visceral disease. The FDA approved trastuzumab deruxtecan for the treatment of HER2-low breast cancer based on DESTINY-Breast04, a randomized, multicenter, open label clinical trial that enrolled 557 adult participants with unresectable or metastatic HER2-low breast cancer. The trial included two cohorts: 494 hormone receptor positive (HR+) participants and 63 hormone receptor negative (HR-) participants. Of these participants, 373 randomly received trastuzumab deruxtecan by intravenous infusion every three weeks and 184 randomly received physician's choice of chemotherapy (eribulin, capecitabine, gemcitabine, nab paclitaxel, or paclitaxel). The results showed improvement in both progression-free survival and overall survival in people with unresectable or metastatic HER2-low breast cancer.

== Management == Genetic mutations of most forms of dwarfism caused by bone dysplasia cannot be altered yet, so therapeutic interventions are typically aimed at preventing or reducing pain or physical disability, increasing adult height, or mitigating psychosocial stresses and enhancing social adaptation. Forms of dwarfism associated with the endocrine system may be treated using hormonal therapy. If the cause is prepubescent hyposecretion of growth hormone, supplemental growth hormone may correct the abnormality. If the receptor for growth hormone is itself affected, the condition may prove harder to treat. Hypothyroidism is another possible cause of dwarfism that can be treated through hormonal therapy. Injections of thyroid hormone can mitigate the effects of the condition, but lack of proportion may be permanent. Pain and disability may be ameliorated by physical therapy, braces or other orthotic devices, or by surgical procedures. The only simple interventions that increase perceived adult height are dress enhancements, such as shoe lifts or hairstyle. Growth hormone is rarely used for shortness caused by bone dysplasias, since the height benefit is typically small (less than 5 cm [2 in]) and the cost high. The most effective means of increasing adult height by several inches is distraction osteogenesis, though availability is limited and the cost is high in terms of money, discomfort, and disruption of life. Most people with dwarfism do not choose this option, and it remains controversial. For other types of dwarfism, surgical treatment is not possible.
=== Food and feed === Microalgae such as Isochrysis galbana, Nannochlor opsisoculata, Chaetoceros muelleri, Chaetoceros gracilis and P. tricornutum have been long utilized in aquaculture as direct or indirect feed sources in hatchery to provide excellent nutritional conditions for early juveniles of farmed fish, shellfish, and shrimp. Furthermore, the EPS layer acts as a nutrient trap, facilitating bacterial growth. The exopolysaccharides of some strains of lactic acid bacteria, e.g., Lactococcus lactis subsp. cremoris, contribute a gelatinous texture to fermented milk products (e.g., Viili), and these polysaccharides are also digestible. An example of the industrial use of exopolysaccharides is the application of dextran in panettone and other breads in the bakery industry. B. subtilis has gained interest for its probiotic properties due to its biofilm which allows it to effectively maintain a favorable microenvironment in the gastrointestinal tract. In order to survive the passage through the upper gastrointestinal tract, B. subtilis produces an extracellular matrix that protects it from stressful environments such as the highly acidic environment in the stomach.
The GoldSrc engine did not provide as much precise control for the player during jumping, making these jumps difficult and often with Freeman falling into a void and the player restarting the game. Wired's Julie Muncy called the Xen sequence "an abbreviated, unpleasant stop on an alien world with bad platforming and a boss fight against what appeared, by all accounts, to be a giant floating infant". The Electric Playground said that Half-Life was an "immersive and engaging entertainment experience" in its first half and that it "peaked too soon". During the AIAS' 2nd Annual Interactive Achievement Awards, Half-Life was awarded "Computer Entertainment Title of the Year" and "PC Action Game of the Year"; it also received nominations for "Game of the Year" and outstanding achievement in "Art/Graphics", "Character or Story Development", "Interactive Design", and "Software Engineering". Jeff Lundrigan reviewed the PlayStation 2 version for Next Generation, rating it three out of five, and wrote that "it may be getting old, but there's still a surprising amount of life in Half-Life". The PlayStation 2 version was a nominee for The Electric Playground's 2001 Blister Awards for "Best Console Shooter Game", but lost to Halo: Combat Evolved for Xbox. In 1999, 2001, and 2005, PC Gamer named Half-Life the best PC game of all time. In 2004, GameSpy readers voted Half-Life the best game of all time. Gamasutra gave it their Quantum Leap Award in the FPS category in 2006. GameSpot inducted Half-Life into their Greatest Games of All Time list in May 2007.

==== Development of new vaccines ==== It is the important mortality in cages among farmed fish, the debates around DNA injection vaccines, although effective, their safety and their side effects but also societal expectations for cleaner fish and security, lead research on new vaccine vectors. Several initiatives are financed by the European Union to develop a rapid and cost-effective approach to using bacteria in feed to make vaccines, in particular thanks to lactic bacteria whose DNA is modified (Boudinot, 2006). In fact, vaccinating farmed fish by injection is time-consuming and costly, so vaccines can be administered orally or by immersion by being added to feed or directly into water. This allows vaccinating many individuals at the same time while limiting the associated handling and stress. Indeed, many tests are necessary because the antigens of the vaccines must be adapted to each species or not present a certain level of variability or they will not have any effect. For example, tests have been done with two species: Lepeophtheirus salmonis (from which the antigens were collected) and Caligus rogercresseyi (which was vaccinated with the antigens), although the homology between the two species is important, the level of variability made the protection ineffective (Fisheries and Oceans Canada, 2014).
Prussia never had more than 320,000 men under arms at any time. In 1813–1815, the core of its army (about 100,000 men) was characterised by competence and determination, but the bulk of its forces consisted of second- and third-line troops, as well as militiamen of variable strength. Many of these troops performed reasonably well and often displayed considerable bravery but lacked the professionalism of their regular counterparts and were not as well equipped. Others were largely unfit for operations, except sieges. During the 1813 campaign, 130,000 men were used in the military operations, with 100,000 effectively participating in the main German campaign, and about 30,000 being used to besiege isolated French garrisons. Spain's armies also peaked at around 200,000 men, not including more than 50,000 guerrillas scattered over Spain. In addition the Maratha Empire, the Ottoman Empire, Italy, Naples and the Duchy of Warsaw each had more than 100,000 men under arms. Even small nations now had armies rivalling the size of the Great Powers' forces of past wars but most of these were poor quality forces only suitable for garrison duties. The size of their combat forces remained modest yet they could still provide a welcome addition to the major powers. The percentage of French troops in the Grande Armée which Napoleon led into Russia was about 50 per cent while the French allies also provided a significant contribution to the French forces in Spain.
== Ureas == Urea (O=C(NH2)2) is more basic at oxygen than simple amides owing to the combined pi-donation from the two amino groups. One consequence is that the inventory of urea complexes is large, including many homoleptic derivatives. Urea forms a broader range of complexes, reflected by the existence of [M(urea)6](ClO4)3 (M = Ti, Mn). As for other complexes of carboxamide ligands, the MOC(NH2)2 core of urea is planar with a bent M-O-C angle. Biuret (H2NC(O)N(H)C(O)NH2) is a derivative of urea but with two amido groups. Biuret forms a variety of metal complexes, e.g. [Cu(H2NC(O)NHC(O)NH2)2]2+. In addition to the parent urea and biuret, many derivatives are known where N-H is replaced by alkyl or aryl.

Purity is normally stated as an area percentage from high-performance liquid chromatography, for example ninety-five or ninety-eight percent. That figure describes the proportion of ultraviolet-absorbing material eluting as the main peak. It says nothing about water content, counterions, residual solvents or mass fraction of the peptide itself.
Peptides are commonly isolated as acetate or trifluoroacetate salts, and the counterion contributes to total mass. Two vials with identical peptide content can therefore differ in weight and in measured response. Residual trifluoroacetate is also relevant in cell-based work because it can influence membrane behaviour.
Degradation products are often closely related to the parent molecule, making them hard to resolve with a single method. Counterion variability adds a further source of scatter between laboratories. Because no pharmacopoeial monograph exists for melanotan-2, groups rely on method-specific validation rather than a shared reference standard.
It is a synthetic cyclic peptide designed as an analog of alpha-melanocyte-stimulating hormone. It acts on melanocortin receptors and is best known from research into pigmentation. It is not an approved pharmaceutical product.
