Melanocortin receptors comprise five subtypes with distinct tissue distributions and functions. Melanotan-2 is described in the literature as a non-selective agonist that engages several of these subtypes, including MC1R, MC3R, MC4R, and MC5R. MC1R is the subtype most directly linked to melanin production in skin cells. Because the compound is not subtype-selective, its observed effects in experimental settings are generally attributed to activity across multiple receptor pathways rather than to a single target.
Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
Lyophilised peptide powder is comparatively stable when kept dry, cold and protected from light. Once dissolved, the molecule is exposed to hydrolysis, oxidation and microbial growth, and degradation accelerates at higher temperatures and in alkaline solution. Repeated freeze-thaw cycles concentrate solutes and promote aggregation. Handling guidance for research peptides commonly clusters around freezer temperatures for powder and short refrigerated use for reconstituted solutions, with pH control and sterile technique applied throughout.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.

The compound was developed in the late 1980s and early 1990s by academic researchers investigating melanocortin signaling and pigmentation. Early work explored whether synthetic analogs could reproduce effects of the natural hormone under controlled conditions. The molecule never advanced through the full regulatory pathway required for approval as a medicine. From the mid-2000s onward it appeared in unregulated consumer markets, often distributed through informal channels. That gap between research origins and commercial availability shapes how the compound is discussed today.
Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
==== Multiple bodies per grave ==== Some couples or groups of people (such as a married couple or other family members) may wish to be buried in the same plot. In some cases, the coffins (or urns) may simply be buried side by side. In others, one casket may be interred above another. If this is planned for in advance, the first casket may be buried more deeply than is the usual practice so that the second casket may be placed over it without disturbing the first. In many states in Australia all graves are designated two or three depth (depending on the water table) for multiple burials, at the discretion of the burial rights holder, with each new interment atop the previous coffin separated by a thin layer of earth. As such all graves are dug to greater depth for the initial burial than the traditional six feet to facilitate this practice. Mass burial is the practice of burying multiple bodies in one location. Civilizations attempting genocide often employ mass burial for victims. However, mass burial may in many cases be the only practical means of dealing with an overwhelming number of human remains, such as those resulting from a natural disaster, an act of terrorism, an epidemic, or an accident. This practice has become less common in the developed world with the advent of genetic testing, but even in the 21st century remains which are unidentifiable by current methods may be buried in a mass grave. Individuals who are buried at the expense of the local authorities and buried in potter's fields may be buried in mass graves.

==== Heart failure ==== Heart failure can be characterized by a loss of β adrenergic receptor (βAR) signalling in heart cells. When the βAR is stimulated by catecholamines such as adrenaline and noradrenaline, there is normally an increase in the contractility of the heart. However, in heart failure there are sustained and elevated levels of catecholamines which result in chronic desensitization of the βAR receptor. This leads to a decrease in the strength of heart contractions. Some research suggests that this chronic desensitization is due to the over activation of a kinase, G protein-coupled receptor kinase 2 (GRK2), which phosphorylates and deactivates certain G protein coupled receptors . When the G protein coupled receptor is activated, the Gβγ subunit recruits GRK2 which then phosphorylates and desensitizes GPCRs like the βAR. Preventing the interaction of the βγ subunit with GRK2 has therefore been studied as a potential target for increasing heart contractile function. The developed molecule GRK2ct is a protein inhibitor which inhibits the signalling properties of Gβγ subunit but does not interfere with alpha subunit signalling. The over expression of GRK2ct has been shown to significantly rescue cardiac function in murine models of heart failure by blocking Gβγ subunit signalling. In another study, biopsies were taken from patients with heart failure and virally induced overexpression of GRK2ct in the heart myocytes. Other tests showed an improvement in cardiac cell contractile function by inhibiting Gβγ.

Tertomotide (GV1001, EARPALLTSRLRFIPK) is a 16-amino acid peptide based on the 616‑626 position fragment of human telomerase reverse transcriptase protein (hTERT). It was originally developed as a possible subunit vaccine for various types of cancer, as it can trigger immune attack against cancer cells which overexpress telomerase, a common mutation found in malignant tumors. However, it does not block telomerase activity in healthy tissue and has been found to produce various useful activities in its own right including antiviral, antiinflammatory and antidepressant effects, and has been trialled for numerous other medical applications including Alzheimer's disease as well as its original role in the treatment of cancer.
=== Physical therapy === Though physical therapy remedies have been scarcely documented, some therapeutic exercises are used to help manage lower back, neck, knee, and shoulder pain. There is moderate quality evidence that therapeutic exercise programs help reduce pain and improve function. Therapeutic exercises include:
The cap is 1.6–8 cm (3⁄4–3+1⁄4 in), conic to convex with a central papilla when young, becoming broadly convex to plane with age, retaining a slight umbo sometimes surrounded by a ring-shaped depression. The cap surface is smooth and sticky, sometimes with white universal veil remnants attached. The cap is brown becoming paler to almost white at the margin and fades to more golden-brown or yellowish with age. When bruised, all parts of the mushroom stain blue. The narrow grey gills are adnate to adnexed, sometimes seceding attachment, and darken to purplish-black and somewhat mottled with age. The gill edges remain whitish. The hollow white stipe is 4–15 cm (2–6 in) high by 0.4–1.4 cm (1⁄4–1⁄2 in) thick, becoming yellowish in age. The well-developed veil leaves a persistent white membranous ring whose surface usually becomes the same color as the gills because of falling spores. The fruiting bodies are 90% water. The mushroom has no odor and has been described as tasting farinaceous, with an alkaline or metallic aftertaste. The spores are 11.5–17.3 x 8–11.5 μm, sub-ellipsoid, basidia 4-spored but sometimes 2- or 3-, pleurocystidia and cheilocystidia present.
==== Beta-blockers ==== Beta-blockers act as competitive antagonists that block the effects of catecholamines at beta-adrenergic receptor sites, resulting in reduced rate and force of contraction of the heart, as well as reduced peripheral vascular resistance.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
Mass spectrometry combined with liquid chromatography provides both molecular weight confirmation and a measure of related impurities. Tandem mass spectrometry adds sequence information. A purity percentage reported without a mass measurement does not confirm what the material is.
