Melanotan II raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
This page was last updated on 2026-04-30 and is reviewed periodically as new material appears.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Outside regulated medicine, melanotan II circulates through online vendors as a research chemical, often marketed for tanning. Products sold this way vary widely in purity, concentration, and labeling accuracy, and independent testing has documented discrepancies. Published reports describe both pigment effects and adverse reactions, including nausea, flushing, and darkening of existing moles. Long-term safety data are sparse, and no large controlled trial has established a risk profile. Questions about cumulative effects on melanocytes remain unresolved in the literature.
Melanotan II is a synthetic peptide analog modeled on alpha-melanocyte-stimulating hormone, a naturally occurring signaling peptide involved in pigmentation. Its structure is a cyclic heptapeptide containing two non-natural substitutions, norleucine at position four and D-phenylalanine at position seven. These modifications resist enzymatic breakdown and extend the molecule's activity relative to the native hormone. The compound binds melanocortin receptors and is studied mainly as a pharmacological tool rather than a therapeutic product. It has never received approval as a medicine in any major jurisdiction.

Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Gram-positive cocci (Staphylococcus aureus, Streptococcus pyogenes, and Enterococci) Gram-negative rods (Escherichia coli, Pseudomonas aeruginosa, Klebsiella species, Bacteroides species, Prevotella species) Clostridium species (Clostridium perfringens, Clostridium septicum, and Clostridium sordellii) In polymicrobial (mixed) infections, Group A Streptococcus (S. pyogenes) is the most commonly found bacterium, followed by S. aureus. However, when the infection is caused solely by S. pyogenes and/or S. aureus, it is classified as a Type II infection. Gram-negative bacteria and anaerobes, such as Clostridia, are more often implicated in Fournier gangrene. This is a subtype of Type I infections affecting the groin and perianal areas. Clostridia account for 10% of overall Type I infections and typically cause a specific kind of necrotizing fasciitis known as gas gangrene or myonecrosis. Type II infection: This infection accounts for 20–30% of cases, mainly involving the extremities. This involves Streptococcus pyogenes, alone or in combination with staphylococcal infections. Methicillin-resistant Staphylococcus aureus (MRSA) is involved in up to a third of Type II infections. Infection by either type of bacteria can progress rapidly and manifest as shock. Type II infection more commonly affects young, healthy adults with a history of injury. Type III infection: Vibrio vulnificus is a bacterium found in saltwater. It occasionally causes NF after entering the body through a break in the skin. One in three patients with a V. vulnificus infection develop necrotizing fasciitis.
The provisional government, with Dupont de l'Eure as its president, consisted of Lamartine for foreign affairs, Crémieux for justice, Ledru-Rollin for the interior, Carnot for public instruction, Goudchaux for finance, Arago for the navy, and Burdeau for war. Garnier-Pagès was mayor of Paris. But, in 1830, the republican-socialist party set up a rival government at the Hôtel de Ville (city hall), including Louis Blanc, Armand Marrast, Ferdinand Flocon, and Alexandre Martin, known as Albert L'Ouvrier ("Albert the Worker"), which bid fair to involve discord and civil war. But this time the Palais Bourbon was not victorious over the Hôtel de Ville. It had to consent to a fusion of the two bodies, in which, however, the predominating elements were the moderate Republicans. It was uncertain what the policy of the new government would be. One party seeing that despite the changes in the last sixty years of all political institutions, the position of the people had not been improved, demanded a reform of society itself, the abolition of the privileged position of property, which they viewed as the only obstacle to equality, and as an emblem hoisted the red flag (the 1791 red flag was, however, the symbol not merely of the French Revolution, but rather of martial law and of order). The other party wished to maintain society on the basis of its traditional institutions, and rallied around the tricolore.
Neanderthal pigmentation genes in modern non-African people are associated with both lighter and darker skin tones, as well as lighter and darker hair colors, suggesting Neanderthals themselves may have had variable pigmentation. Variants associated with red hair have been found in some Neanderthals, but they do not appear to have been common. Neanderthal skin and hair pigmentation variants are under positive selection in non-Africans, unlike most other Neanderthal genes, suggesting that the adaptive value of lighter features was beneficial for modern humans in Eurasia, where there is significantly less sunlight as compared to Africa.
Pan pizza, baked in a pan with a crispy edge; "Stuffed crust" pizza, with the outermost edge wrapped around a cylinder of mozzarella cheese; "Hand-tossed", more like traditional pizzeria crusts; "Thin 'N Crispy", a thin, crisp dough which was Pizza Hut's original style; Dippin' Strips pizza, a pizza cut into small strips that can be dipped into a number of sauces; The P'Zone, a pizza-calzone hybrid with a marinara dipping sauce that comes in plain, Supremo, Meaty, and pepperoni; The Bigfoot pizza, its largest product; The Priazzo, a pie like pizza stuffed with pizza ingredients. The "stuffed crust" pizza was introduced on March 26, 1995. By the end of the year, it had become one of their most popular lines.
=== Climate === Lower Saxony falls climatically into the north temperate zone of central Europe that is affected by prevailing Westerlies and is located in a transition zone between the maritime climate of Western Europe and the continental climate of Eastern Europe. This transition is clearly noticeable within the state: while the northwest experiences an Atlantic (North Sea coastal) to Sub-Atlantic climate, with comparatively low variations in temperature during the course of the year and a surplus water budget, the climate towards the southeast is increasingly affected by the Continent. This is clearly shown by greater temperature variations between the summer and winter halves of the year and in lower and more variable amounts of precipitation across the year. This sub-continental effect is most sharply seen in the Wendland, in the Weser Uplands (Hamelin to Göttingen) and in the area of Helmstedt. The highest levels of precipitation are experienced in the Harz because the Lower Saxon part forms the windward side of this mountain range against which orographic rain falls. The average annual temperature is 8 °C (46 °F); 7.5 °C (45.5 °F) in the Altes Land and 8.5 °C (47.3 °F) in the district of Cloppenburg.

William Field and colleagues found a 50% increased lung cancer risk even at the protracted exposures at the EPA's action level of 4 pCi/L. North American and European pooled analyses further support these findings. However, the conclusion that exposure to low levels of radon leads to elevated risk of lung cancer has been disputed, and analyses of the literature point towards elevated risk only when radon accumulates indoors and at levels above 100 Bq/m3. Thoron (220Rn) is less studied than 222Rn in regards to domestic exposure due to its shorter half-life. However, it has been measured at comparatively high concentrations in buildings with earthen architecture, such as traditional half-timbered houses and modern houses with clay wall finishes, and in regions with thorium- and monazite-rich soil and sand. Thoron is a minor contributor to the overall radiation dose received due to indoor radon exposure, and can interfere with 222Rn measurements when not taken into account.
=== Intracrines and regenerative Medicine === The discovery of intracrine loops in stem cell regulation has profound implications for regenerative medicine. Because intracrines can establish long-lasting differentiation programs, they offer potential therapeutic targets for tissue regeneration and repair. For example, in cardiac repair, HMGB1 has been shown to enhance the proliferation and differentiation of cardiac stem cells following myocardial infarction, suggesting that modulating intracrine pathways could improve heart regeneration. The ability of certain intracrines to reprogram cells into pluripotent-like states also opens new avenues for regenerative therapies. Oct3/4, Sox2, and Nanog, all of which are involved in maintaining stem cell pluripotency, can potentially be introduced into cells to drive reprogramming without the need for genetic modification. This approach could provide safer and more controlled methods for generating patient-specific stem cells.
And here is The Mystery of the Yellow Room. That - ah, that is really a classic! I approve of it from start to finish. Such a logical approach! There were criticisms of it, I remember, which said it was unfair. But it is not unfair, my dear Colin. No, no. Very nearly so, perhaps, but no not quite. There is the hair's breadth of difference. No. All through there is truth, concealed with a careful and cunning use of words. Everything should be clear at that supreme moment when the men meet at the angle of the three corridors." He laid it down reverently. "Definitely a masterpiece, and, I gather, almost forgotten nowadays. In a 1981 poll by Edward D. Hoch of 17 mystery writers and reviewers, this novel was voted the third-best locked-room mystery of all time, behind Hake Talbot's Rim of the Pit (1944) and John Dickson Carr's The Hollow Man (1935). The popular 1946 Japanese detective novel The Honjin Murders (本陣殺人事件, Honjin satsujin jiken) by Seishi Yokomizo refers to The Mystery of the Yellow Room numerous times, the narrator quipping that Leroux's novel "bears the closest resemblance" to the story recounted in the novel. The final episode of the 2025 Netflix miniseries The Residence is titled The Mystery of the Yellow Room and concludes the series with a locked-room murder deduction.
==== Steatosis ==== Hepatotoxicity may manifest as triglyceride accumulation, which leads to either small-droplet (microvesicular) or large-droplet (macrovesicular) fatty liver. There is a separate type of steatosis by which phospholipid accumulation leads to a pattern similar to the diseases with inherited phospholipid metabolism defects (e.g., Tay–Sachs disease)

== Third party payment == Most insurers refuse to cover the treatment. In some hearings and cases where a judge has heard evidence, insurance companies have been ordered to pay for that patient. For example, a trial with CalPERS resulted in a decision ordering Blue Cross and other insurance providers to pay for the therapy as to those parties. However, a subsequent assessment by CMS found no evidence that Pulsatile Insulin improves the condition of type 1 and 2 diabetics and has issued a National non-coverage decision which to date is still in effect. CMS has issued a specific code to use when billing this treatment to avoid erroneous payment by billing by the individual procedures that are used in the treatment. Until acceptable clinical trials are performed to show the benefit of pulsatile insulin or Artificial Pancreas Treatment as it is being currently marketed as, the Medicare NCD will continue to remain in effect.

In February 2005, Bagwell and Biggio were jointly inducted into the Texas Sports Hall of Fame. Shortly after the 2005 season began, the chronic arthritic condition in his shoulder that had begun in 2001 finally sidelined him, rendering him inactive for three-quarters of the season. The former Gold Glove winner was now a defensive liability with a severely limited throwing arm; he had to "push" the ball instead of throwing it. Teams began taking advantage of his defensive weakness. Once possessing great ability to throw out the lead runner at third base ahead on bunt plays, Bagwell found it difficult to practice with the other infielders between innings. Concurrently, Bagwell's offensive production suffered, and pressure mounted on Astros management to bench the perennial All-Star; the club had started the season with a 15–30 won–loss record. He hit his last major league home run against Maddux on April 29, tying him for the most against any pitcher with seven. Bagwell continued to play through the pain until, after going 0-for-5 in a loss to the Pirates on May 4, it became so unbearable that he asked manager Phil Garner to remove him from the lineup the following day. He had hit just .250 with three home runs in 88 at-bats. The Astros placed him on the disabled list a few days later, and shoulder surgery followed. The Astros dramatically improved after their poor start to finish 74–43 over their final 117 games and capture the NL wild card.
=== Ocular myxoma === Myxoma is a rare, benign stromal tumor of mesenchymal origin often confused with other conjunctival stromal tumors. Conjunctival myxomas are thought to originate in Tenon's capsule and can masquerade as conjunctival lymphoma, lymphangioma, ocular surface squamous neoplasia (OSSN), or amelanotic melanoma.
The gamma decay from a metastable state is referred to as isomeric transition (IT), or internal transition, though it resembles shorter-lived "prompt" gamma decays in all external aspects with the exception of the longer life. This is generally associated with a high nuclear spin change, or "forbiddenness", which would be required in gamma emission to reach the ground state; this is even more true of beta decays. A low transition energy both slows the transition rate and makes it more likely that only highly forbidden decays are available, so most long-lived isomers have a relatively low excitation energy above the ground state (in the extreme case of thorium-229m, low excitation alone causes the measurably long life). In 210m83Bi, the forbiddenness of available beta and gamma decays is so high that alpha decay is observed exclusively, though even that is slower than for the ground state. For most lighter isomers including 180m73Ta, alpha decay is not practically available, but others are not quite so forbidden as those two. The first nuclear isomer and decay-daughter system (uranium X2/uranium Z, now known as 234m91Pa/23491Pa) was discovered by Otto Hahn in 1921. Metastable isomers can be produced through any nuclear reaction, including radioactive decay, neutron capture, nuclear fission, and bombardment by accelerated charged particles. A nucleus produced this way generally starts its existence in an excited state that loses its excess energy through the emission of one or more gamma rays or conversion electrons.

No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
Afamelanotide is the linear analogue [Nle4-D-Phe7]-alpha-MSH, sometimes called melanotan I, while melanotan II is cyclic and carries three further substitutions. The two are distinct molecules and are not interchangeable in analytical testing.
Binding to the four melanocortin receptor subtypes is well documented in vitro. The relative contribution of each subtype to whole-body effects in humans is far less certain.
It is a synthetic cyclic peptide designed as an analog of alpha-melanocyte-stimulating hormone. It acts on melanocortin receptors and is best known from research into pigmentation. It is not an approved pharmaceutical product.
