Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Because the substance circulates mainly through informal markets, verification is a recurring theme in technical discussion. Independent analyses have found that labeled content and actual content can diverge, and that purity varies between samples. Analytical laboratories use reversed-phase chromatography to separate components and mass spectrometry to confirm identity. Isotope-labeled internal standards improve quantification in complex matrices. Such methods describe what a sample contains but say nothing about its sterility, lawful status, or suitability for any use. Open questions remain about how consistently testing is applied across the supply chain.
Regulatory treatment of this peptide varies by country. It holds no marketing authorization as a medicine in the United States, the European Union, or most other jurisdictions. Some countries classify products containing it as prescription-only or unlicensed medicines, which restricts lawful supply. Authorities have issued public notices warning that unregulated products may contain undeclared or incorrect ingredients. The molecule also appears on prohibited lists for competitive sport. These measures address supply oversight rather than any approved therapeutic role.

Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Melanotan-2 appears on the World Anti-Doping Agency prohibited list within the peptide hormone class, and several national regulators treat it as an unapproved prescription substance. Some countries restrict importation or sale for personal use. Because the compound is widely traded as a research chemical, the practical legal picture differs between jurisdictions and shifts over time. Human safety data covering long periods are limited, and whether repeated pigmentation changes carry any lasting risk to melanocytes remains an open question.
Freeze-dried melanotan-2 is normally kept as a desiccated powder at minus twenty degrees Celsius or lower, shielded from light and moisture. Peptides of this size degrade through hydrolysis, oxidation and deamidation, and each pathway accelerates as temperature and water activity rise. Repeated freeze-thaw cycles promote aggregation and loss of material, so aliquoting a stock solution before freezing is standard laboratory practice. Once dissolved, the solution is markedly less stable than the powder. In laboratory work, solutions are generally refrigerated and used within days rather than kept for months.
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography, which separates the target peptide from truncated or oxidised impurities. Mass spectrometry, most often coupled to liquid chromatography, confirms molecular mass and detects substitutions that chromatography alone may miss. Amino acid analysis and peptide mapping supply additional structural evidence, while nuclear magnetic resonance is reserved for full structural confirmation. Laboratories that examine samples sold online report wide variation in actual content, with some vials containing little or none of the labelled material.

Cloning of the YAP1 gene facilitated the identification of a modular protein domain, known as the WW domain. Two splice isoforms of the YAP1 gene product were initially identified, named YAP1-1 and YAP1-2, which differed by the presence of an extra 38 amino acids that encoded the WW domain. Apart from the WW domain, the modular structure of YAP1 contains a proline-rich region at the very amino terminus, which is followed by a TID (TEAD transcription factor interacting domain). Next, following a single WW domain, which is present in the YAP1-1 isoform, and two WW domains, which are present in the YAP1-2 isoform, there is the SH3-BM (Src Homology 3 binding motif). Following the SH3-BM is a TAD (transactivation domain) and a PDZ domain-binding motif (PDZ-BM) (Figure 1).
=== SIMBAS === In March 2011, a team of researchers from UC Berkeley, DCU and University of Valparaíso have developed lab-on-a-chip that can diagnose diseases within 10 minutes without the use of external tubing and extra components. It is called Self-powered Integrated Microfluidic Blood Analysis System (SIMBAS). It uses tiny trenches to separate blood cells from plasma (99 percent of blood cells were captured during experiments). Researchers used plastic components, to reduce manufacturing costs.

==== Absorption ==== Following a single oral dose of sertraline, mean peak blood levels of sertraline occur between 4.5 and 8.4 hours. Bioavailability is likely linear and dose-proportional over a dose range of 150 to 200 mg. Concomitant intake of sertraline with food slightly increases sertraline peak levels and total exposure. There is an approximate 2-fold accumulation of sertraline with continuous administration and steady-state levels are reached within one week.
=== Zostavax === The live vaccine (Zostavax) is very safe; one to a few percent of people develop a mild form of chickenpox, often with about five or six blisters around the injection site, and without fever. The blisters are harmless and temporary. In one study 64% of the Zostavax group and 14% of the controls had some adverse reaction. However, the rates of serious adverse events were comparable between the Zostavax group (0.6%) and those receiving the placebo (0.5%). A study including children with leukaemia found that the risk of getting shingles after vaccination is much lower than the risk of getting shingles for children with natural chicken pox in their history. Data from healthy children and adults point in the same direction. Zostavax is not used in people with compromised immune function.
Antimicrobial stewardship programmes appear useful in reducing rates of antimicrobial resistance. The antimicrobial stewardship program will also provide pharmacists with the knowledge to educate patients that antibiotics will not work for a virus for example. Excessive antimicrobial use has become one of the top contributors to the evolution of antimicrobial resistance. Since the beginning of the antimicrobial era, antimicrobials have been used to treat a wide range of infectious diseases. Overuse of antimicrobials has become the primary cause of rising levels of antimicrobial resistance. Prescribers are willing to prescribe antimicrobials to individuals who believe that antimicrobials can cure nearly all illnesses, including viral infections like the common cold. In an analysis of drug prescriptions, 36% of individuals with a cold or an upper respiratory infection (both usually viral in origin) were given prescriptions for antibiotics. These prescriptions increase the risk of further evolution of antibiotic resistant bacteria. Using antimicrobials without prescription is another driving force leading to the overuse of antibiotics to self-treat diseases like the common cold, cough, fever, and dysentery resulting in an epidemic of antibiotic resistance in countries like Bangladesh, risking its spread around the globe. Introducing strict antibiotic stewardship in the outpatient setting to reduce inappropriate prescribing of antibiotics may reduce the emerging bacterial resistance.
=== Ecology === Naegleria fowleri, an excavate, inhabits soil and water. It is sensitive to drying and acidic conditions, and cannot survive in seawater. The amoeba thrives at moderately elevated temperatures, making infections more likely during the summer months. N. fowleri is a facultative thermophile, capable of growing at temperatures up to 46 °C (115 °F). Warm, fresh water with an ample supply of bacteria as food provides a suitable habitat for amoebae. Locations where many amoebic infections have occurred include artificial bodies of water, disturbed natural habitats, areas with soil, and unchlorinated or unfiltered water. N. fowleri appears to flourish during periods of disturbance. The "flagellate-empty" hypothesis suggests that N. fowleri's success may stem from decreased competition when thermosensitive protozoal fauna do not survive temperature changes. In other words, N. fowleri thrives when other predators consuming its food supply are absent. This hypothesis implies that human disturbances, such as thermal pollution, increase the abundance of N. fowleri by eliminating its resource competitors. Amoeboflagellates have a motile, flagellated stage that aids in dispersal, which is advantageous in environments cleared of competing organisms.

== International Cooperation == NCCD participates in global activities on matters of agri-logistics and food loss and waste. In 2013, Capt. Pawanexh Kohli, then head of NCCD, represented India as the Guest of Honour at the International Summit on Sustainability and cold chain in France, when NCCD also signed an MoU with Cemafroid. NCCD's first CEO was also invited for a debate at UK's House of Lords in 2015 and as expert to UK's Policy Commission on Cold Economy. That same year, NCCD's CEO co-chaired Working Groups on Food Loss and Food Waste at the International Action Summit held in The Hague. Under the authorship of its founding CEO, NCCD published India's minimum system standards and guidelines for cold chain infrastructure. NCCD also conducted the country's first ever cold chain infrastructure assessment, which thereafter formed the basis for future strategies in India. In 2016, NCCD was designated a knowledge partner of India's inter-ministerial Committee for Doubling Farmers' Income, with its CEO as a member of this historic Committee. NCCD has undertaken projects to demonstrate the efficiency of cold chain in connecting farmers with markets across India's sub-continental distances. Such projects also documented higher incomes for farmers and lower food losses and carbon footprint of the supply chain. Another initiative by NCCD is that of bringing synergistic uses to village level refrigerated pack-houses so as to benefit social and welfare needs of rural communities.
The two amphetamine enantiomers that compose Adderall, such as Adderall tablets/capsules (levoamphetamine and dextroamphetamine), alleviate the symptoms of ADHD and narcolepsy by increasing the activity of the neurotransmitters norepinephrine and dopamine in the brain, which results in part from their interactions with human trace amine-associated receptor 1 (hTAAR1) and vesicular monoamine transporter 2 (VMAT2) in neurons. Dextroamphetamine is a more potent CNS stimulant than levoamphetamine, but levoamphetamine has slightly stronger cardiovascular and peripheral effects and a longer elimination half-life than dextroamphetamine. The active ingredient in Adderall, amphetamine, shares many chemical and pharmacological properties with the human trace amines, particularly phenethylamine and N-methylphenethylamine, the latter of which is a positional isomer of amphetamine. In 2023, Adderall was the fifteenth most commonly prescribed medication in the United States, with more than 32 million prescriptions.

Nucleotides are made from amino acids, carbon dioxide and formic acid in pathways that require large amounts of metabolic energy. Consequently, most organisms have efficient systems to salvage preformed nucleotides. Purines are synthesized as nucleosides (bases attached to ribose). Both adenine and guanine are made from the precursor nucleoside inosine monophosphate, which is synthesized using atoms from the amino acids glycine, glutamine, and aspartic acid, as well as formate transferred from the coenzyme tetrahydrofolate. Pyrimidines, on the other hand, are synthesized from the base orotate, which is formed from glutamine and aspartate.
== Developmental history == ZooMS was first published in 2009 by a team of researchers from the University of York, but the term was coined later in a publication in 2010. The original aim of ZooMS was to distinguish between sheep and goat. The bones of these two closely related species are difficult to distinguish, especially when fragmented, yet the difference between these two common domesticates is very important for our understanding of past husbandry practices. Most of the method development following the initial publication of ZooMS has focused on the extraction of collagen from the archaeological material. In the original protocol acid was used to dissolve the bone's mineral matrix and free up the collagen. In 2011 an alternative extraction method was published that used an ammonium bicarbonate buffer to solubilise the collagen without dissolving the mineral matrix. In contrast to the acid protocol, the ammonium bicarbonate protocol does not affect the size and mass of the sample, making it a much less destructive method compared to the original protocol. In fact, the ammonium bicarbonate protocol was proposed as a non-destructive protocol for ZooMS, but in practice destructive samples are still taken for this protocol (see ). Submerging a sample in ammonium bicarbonate does chemically alter the sample, which is why current practices continue to take a destructive sample.

No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
Afamelanotide is the linear analogue [Nle4-D-Phe7]-alpha-MSH, sometimes called melanotan I, while melanotan II is cyclic and carries three further substitutions. The two are distinct molecules and are not interchangeable in analytical testing.
Binding to the four melanocortin receptor subtypes is well documented in vitro. The relative contribution of each subtype to whole-body effects in humans is far less certain.
Only a small number of early-stage human studies have been reported, and most were limited in size and duration. No large late-stage program has established a general efficacy or safety profile. The evidence base is therefore thin compared with approved medicines.
